Function/Usage

Synthetic manganese-porphyrin complexes have been documented to act as scavengers for oxidative species such as peroxynitrite, superoxide and hydrogen peroxide. EUK 134 is a salen-manganese complex that has been modified to increase its catalase activity while retaining SOD activity.

EUK 134 has potent antioxidant activity and inhibits β-amyloid (β-amyloid) and related amyloid fibril formation.

EUK134 inhibits membrane damage-induced activation of the mitogen-activated protein kinase pathway, and reduces p53 accumulation in UV-B-irradiated primary human keratinocytes. EUK134 (20 μM) prevents Aβ-induced microglia proliferation in vitro. In SK-N-MC cells, EUK134 attenuates oxidative stress by inhibiting MAPK channels, protects neuronal cells from H(2)O(2) toxicity, and causes the expression of pro-apoptotic genes p53 and Bax to decrease, while also enhancing The expression of anti-apoptotic Bcl-2 gene. EUK134 significantly inhibited amyloid formation at two molar ratios of 1:1 and 5:1 (drug to protein).
